- Abstract: Lignocellulosic materials, due to their significant potential in innumerous applications, have been extensively studied in the last three decades. Corn cob as a lignocellulosic feedstock offers promising possibilities for bioproducts production. Through pretreatments, enzymatic hydrolysis and fermentation, the biomass may be converted to bioproducts. Those bioproducts may be a green alternative to various petrochemicals. Twin screw extrusion process is an alternative pretreatment method due to a variety of advantages, such as high solids loading and continuous process. The aim of this work was to evaluate the feasibility of employing extruded corn cob on the enzymatic process, increasing the solids loading of biomass. The dosage of enzymes and the solids loading were optimized by response surface methodology (RSM) to obtain high values of cellulose conversion to glucose, and glucose productivity. The RSM resulted in an increase in the solids loading of 78%, to 17,8 %w/w by using an enzyme loading of 31 FPU/g, achieving a glucose productivity of 0.69 gL-1h-1 and a cellulose conversion to glucose of 91.84 %w/w. A kinetic evaluation of the enzymatic hydrolysis process leads to a Km of 22.59 gL-1 and a maximum velocity of cellulose conversion to glucose of 6.0 %w/w h-1.
