Thermothelomyces thermophilus as industrial cell factory for high titter of biomoleculaes production (2023)

- Abstract: Filamentous fungi naturally secrete a diverse array of proteins and enzymes to decompose various organic carbon sources (e.g., lignocellulosic biomass) into monomeric sugars (e.g.,glucose and xylose) for their metabolic processes. These fungi can serve as excellent industrial platforms for the production of bioproducts with biotechnological significance, owing to their ability to achieve high secretion titers of biomolecules, their easy industrial manipulation, and their capacity to perform various post-translational modifications, such as glycosylation and disulfide bond formation, unlike observed in other industrial cell factories, like bacteria and yeast. Among these, Thermothelomyces thermophilus emerges as a particularly promising industrial cell factory for biomolecules production, displaying morphological characteristics suitable for large-scale fermentable processes. To utilize T. thermophilus as a cell factory for protein production, we optimized the cultivation conditions and carbon sources in submerged fermentation. The submerged fermentations were conducted in 250 ml flasks, at controlled temperature and agitation, employing three different carbon sources (glucose, lactose, and sucrose) and two distinct media (Minimal and Mandels-Weber). The consumption of these carbon sources, pH variation, extracellular protein production, and fungi biomass were evaluated. The results allowed us to identify the optimal media, carbon source, and culture conditions for producing high levels of extracellular proteins from T. thermophilus. By defining cultivation parameters and the profile of carbon source consumption, we can improve T. thermophilus as an industrial cell factory through synthetic modifications aiming to increase extracellular protein production in submerged fermentation processes.
