Interneuronsand medium spiny in the nucleus accumbens of the rat express distinct patterns of ampa-type receptor subunits. (2008)

- Abstract: Objectives: The nucleus accumbens (Acb) is a key structure of the reward system and crucially involved in different aspects of adaptative and emotional behaviors. The great majority of neurons in the Acb are medium spiny projection neurons (MSNs) that are modulated by different types of interneurons. There is little information which AMPA-type receptor subunits are expressed by MSNs and interneurons of the Acb. Thus, we investigated the distribution of all AMPA-type receptor subunits in the Acb and their co-localization with the neuronal phosphoprotein DARPP-32, which is expressed in all MSNs expressing dopamine receptors, as well as with markers selectively expressed in interneurons such as parvalbumin and Chat. Methods: Adult Wistar rats (n= 40) were perfused with 4% paraformaldehyde and brains were cut on a freezing microtome. Sections through the Acb were either incubated with antibodies to the AMPA-type receptor subunits GluR1, GluR2, GluR2/3, and GluR4 alone, or together with antibodies to DARPP-32, parvalbumin, Chat and the neuronal marker NeuN.Standard immunoperoxidase or fluorescence techniques were used for visualization. Results: The Glur2/3 and GluR2 subunits showed by far the strongest expression and were expressed in all DARPP-32+ neurons. Double-staining with NeuN revealed that the GluR2 subunit was expressed in almost all neurons of the Acb, except in cholinergic Chat+ interneurons. In contrast, the GluR1 and GluR4 subunits were only weaklyexpressed in the Acb and often co-localized with parvalbumin. Conclusions: Results indicate that 1.) all MSNs receive converging glutamatergic and dopaminergic inputs; 2.) the GluR2 subunit might play a prominent role in mediating dopamine - glutamate interactions; and 3.) MSNs and interneurons in the Acb express distinct patterns of AMPA-type subunits, with the GluR1 and GluR4 subunits exclusively expressed in interneurons.
