Distribution of the serotonin 2A receptor in the prefrontal cortex and mesencephalon of the rat and its relation to the dopaminergic system (2006)

- Abstract: Objetivo: Serotonin 2A (5-HT2A) receptors are supposed to play an essential role in mediating serotonin-dopamine interactions. To clarify the anatomical relationships of this receptor type with the dopaminergic system, we investigated 1.) its distribution in the prefrontal cortex (PFC), substantia nigra (SN), and ventral tegmental area (VTA) by single label immunohistochemistry and 2.) its relation to dopaminoceptive neurons in the PFC and dopaminergic neurons in the mesencephalon by double-label immunohistochemistry. Métodos e Resultados: Adult rats (Wistar, n = 8) were perfused with 4% paraformaldehyde. Brains were cut on a vibratome and coronal sections through the PFC and mesencephalon were either incubated with a polyclonal antibody to 5-HT2A alone or together with monoclonal antibodies to DARPP-32 or tyrosine hidroxylase (TH). Standard immunoperoxidase or fluorescence techniques were used for visualization.As revealed by single labeling, intense 5-HT2A labeling could be observed in pyramidal cells in layer III and in superficial layer V of the PFC, with their principal dendrites typically stained in their whole extension. In the SN and VTA, several neuronal somata were labeled for 5-HT2A. Double labeling for 5HT2A and TH indicated that almost all 5-HT2A+ neurons in the SN and VTA also express TH. Double labeling for 5HT2A and DARPP-32 in the PFC demonstrated that these markers are mainly expressed in 2 different populations of pyramidal cells.Conclusões: Our results indicate that 5-HT2A receptors are strongly expressed in the PFC. Although 5-HT2A receptors and DARPP-32 seem to be expressed in two different populations of pyramidal cells in the PFC, the fact that a great number of TH+ cells in the SN and VTA are double-labeled for 5-HT2A indicates that serotonin might modulate dopaminergic functions in the PFC by activating 5-HT2A receptors in the mesencephalon
