Distribution of AMPA and NMDA type glutamate receptors and their relation to the dopaminergic system in the nucleus accumbens of the rat (2006)

- Abstract: Objetivo: The nucleus accumbens (Acb) is a key structure of the reward system of the brain and crucially involved in appetitive behaviors. There is increasing evidence that dopaminergic and glutamatergic afferents converge on the same target cells in the Acb. In order to determine the anatomical relationships of AMPA and NMDA-type glutamate receptors with the dopaminergic system, we 1.) investigated their distribution within the Acb by single-labeling and 2.) their co-localization with markers of the dopaminergic system such as tyrosine hydroxylase (TH), DARPP-32, and D2 receptors by double-label immunohistochemistry. Métodos e Resultados: Adult Wistar rats (n = 24) were perfused with 4% paraformaldehyde and brains were cut on a vibratome. Coronal sections through the Acb were either incubated with antibodies to the AMPA type receptor subunits GluR1, GluR2/3, GluR4, or to the NMDAR1 receptor alone, or together with antibodies to TH, DARPP-32 or the D2 receptor. Standard immunoperoxidase or fluorescence techniques were used for visualization. As revealed by single labeling, GluR1, GluR2/3, GluR4, and NMDA receptor were homogeneously expressed in the core and shell of the Acb. Among the AMPA type receptor subunits, the GluR2/3 receptor subunit showed by far the strongest expression. As revealed by double-label immunohistochemistry, all glutamate receptor subtypes examined were to a certain degree co-localized with DARPP-32+ neurons in the Acb, with the GluR2/3receptor showing the highest degree of co-localization. Conclusões: Our findings indicate that all major glutamate receptor subtypes in the Acb are closely related to markers of the dopaminergic system and that the AMPA receptor subunit GluR2/3 might play a pivotal role in mediating dopamine - glutamate interactions
