Dopamine -glutamate interactions in DARPP-32 positive neurons of the prefrontal cortex of the rat are partly mediated by the AMPA type receptor subunit GLUR2/3 (2006)

- Abstract: There is increasing evidence that dopaminergic and glutamatergic afferents converge on the same target neurons in the prefrontal cortex (PFC) and that dopamine (DA) acts as a neuromodulator that alters the postsynaptic cell's response to gluamatergic inputs in a manner that is highly dependant on the DA and glutamate (GLU) receptor types involved. The neuronal phosphoprotein DARPP-32 has been proposed to be a major intracellular integrator of DA - GLU interactions. In order to determine the role of AMPA receptors in mediating DA - GLU interactions in the medial PFC of the rat, we investigated by single and double-label immunohistochemistry the distribution and co-localization with DARPP-32 of the AMPA type receptor subunit GluR2/3. Adult rats (Wistar, n = 24) were perfused with 4% paraformaldehyde and brains were cut on a vibratome. Coronal sections through the PFC were either incubated with a polyclonal antibody to the GluR2/3 receptor alone or together with a monoclonal antibody to DARPP-32. Standard immunoperoxidase or fluorescence techniques were used for visualization.As revealed by single label immunohistochemistry, intense GluR2/3 labeling could be observed in neuronal somata in layer II and in pyramidal cells of superficial layers III and V of the PFC. In addition, weak GluR2/3 labeling could be found in layer VI. DARPP-32+ neurons were generally highly enriched in cortical layer VI and moderately expressed in layers II, III, and V. Asrevealed by double labeling for DARPP-32 and GluR2/3, almost all, if not all, of the DARPP-32+ neurons in the PFC showed at least weak expression of the GluR2/3receptor. These findings indicate that DARPP-32 is co-expressed with the AMPA receptor subunit GluR2/3 in the medial PFC and that this subunit might play a prominent role in mediating DA -GLU interactions
