Initial peptide content characterization in mammalian tissues using LC-ESI-MS/MS (2005)

- Abstract: Objectives Proteasomal proteolyses produces large quantities of intracellular peptides from natural misfolded or aged proteins. Some evidences suggest the involvement of such intracellular peptides as modulators of protein interactions. Therefore, we developed a new methodology that uses inactive oligopeptidases to identify specific peptides in complex biological samples. The goal of the present work was to use LC-ESI-MS/MS to identify and sequence these complex peptide mixtures obtained from filtrates of rodentia tissues.Methods and Results Briefly, tissues were homogenized in 0,05 M HCl and centrifuged at 100.000 x g for 60 min, at 4 C. The supernatants were subjected to size exclusion using a Millipore filter device (Millipore, USA). The resulting material containing peptides smallest than 5 kDa were concentrated using a 1 cc OASIS HLB (Waters, UK) and filtrated using the inactive oligopeptidases. LC-ESI-MS/MS analysis were performed at the Mass Spectrometry Laboratory, National Laboratory of Light Sincrotron (LNLS), Campinas, SP, in a Q-Tof (Waters, UK) coupled to a capillary liquid chromatography system (CapLC Waters). Thirty nine peptides were sequenced in samples from adipose (rat and mouse), brain and spleen (rat) tissues, and match internal regions of known and unknown putative intracellular proteins. Organizing these peptides based on their putative motifs we found that 33% contain protein kinase (PK) C; 23% PKA; 10% C kinase 2 (CK2); 3% C kinase 1 (CK1);05% P38 mitogenactivating PK (MAPK) phosphorylation sites, and 3% contained a glycosilation site. The remaining 9 peptides contained no putative motifs. Conclusion These initial results suggest that most of peptides identified herein contain phosphorylation sites and could be modulating intracellular signaling
