14.3.3 plays important role on endopeptidase 24.15 (EC 3.4.24.25) secretion (2003)

- Abstract: The mechanism responsible for the neuropeptide-metabolizing enzyme endopeptidase 24.15 (EC 3.4.24.15; ep24.15) secretion is unknown, as the protein is encoded without a signal peptide sequence for entering the cell secretory pathway. Herein we describe an increased ep24.15 stimulated secretion upon overexpression of the 14.3.3 protein in human embryoninc kidney 293 cells (HEK293). Protein kinase (PK) A phosphorylation of ep24.15 Ser644 residue was found to be important for direct interaction with 14.3.3. Moreover, ep24.15 S644A mutation ablates the positive secretory effects of 14.3.3 overexpression. Pharmacological analyses shown that forskolin (10 µM) or phorbol ester (80 nM) increases the A23187 (7,5 µM) stimulatory effects of ep24.15 secretion; no such synergism was observed upon PKA or PKC inhibition. The ep24.15 S644A mutation also prevents the synergism between A23187 and PKA or PKC activation. Double labeling confocal microscopy analyses seems to confirm the in vivo association of ep24.15 and 14.3.3 induced by PKA or PKC activation.Therefore, these data suggest that 14.3.3-ep24.15 interactions play a key role on the mechanism of ep24.15 unconventional secretion
