Synthesis of 2-ethylhexyl oleate catalyzed by lipase immobilized on a magnetic support in packed-bed bioreactor (2019)

- Abstract: This study evaluated the synthesis of the emollient ester 2-ethylhexyl oleate catalyzed by Candida antarctica lipase immobilized on a poly(styrene-co-divinylbenzene) support containing magnetite particles. Esterification reactions were conducted in a continuous-flow packed-bed bioreactor and the influence of acid/alcohol molar ratio (1:4 to 4:1) and space- time (3 to 12 h) were evaluated. We observed that the longer the space time and the higher the concentration of either acid or alcohol, the higher the esterification yield (80–85%). These results can be explained by Le Châtelier’s principle: an increase in reagent concentration shifts the equilibrium toward product formation. However, higher productivities were obtained using short space- times and acid/alcohol molar ratios closer to the stoichiometric ratio, because high concentration of the limiting reactant, combined with lower amounts of water and products and shorter space- time, resulted in increased reaction rates. At a space time of 12 h, productivity was highest (22–26 mmol g−1 L−1 h−1) at an acid/alcohol molar ratio of 1:1. The biocatalyst showed good stability (half-life of 2063 h) under these conditions. The results suggest that the bioprocess is attractive for the synthesis of emollient esters.
