Genetic analysis of the oligopeptide binding protein OppA encoded by species of Xanthomonas (2007)

- Abstract: The Xanthomonas genus encompasses a ubiquitous group of plant pathogens, infecting at least 398 species, including plants of major economic interest, such as rice, beans, orange, grape and soy, among others. Genome sequencing projects have highlighted the multiplicity of ABC transporters, responsible for the uptake of nutrients and secretion of secondary metabolytes. The present work investigated the presence of the oppA gene, coding for the homonymous binding protein of the Opp system, in several Xanthomonas species. The Opp system is functional and conserved among a considerable number of other proteobacteria. Surprisingly, southern-blot and immuno-blot experiments could only detect oppA in two Xanthomonas axonopodis citri (Xac) strains (strains 306 and 381), among the Xanthomonas species tested. Selection of a Xac 306 oppA mutant revealed no impairment of small peptides import, and no difference in the degree of virulence when compared to the parental strain. These results suggest a recent evolutionary origin of the Opp system in the Xanthomonas genus, and also points to the possibility of an alternative oligopeptide transport system in these bacteria.
