Analysis of Caulobacter crescentus mutations in response to UV irradiation and methyl methanesulfonate (2005)

- Abstract: The mechanisms of DNA repair are key processes for the maintenance of the genome integrity in every living organism. As the DNA molecule has all the necessary information for the biological system to work, it is comprehensible that its integrity is so necessary for life maintenance. The model organism used nowadays, the bacterium Escherichia coli, does not answer all questions concerning DNA repair processes in bacteria. On the other hand, Caulobacter crescentus is widely employed for studies of cellular differentiation and cell cycle in bacteria. Each cell division produces two different cell types: a motile - flagellum-and a stalked, sessile cell. Both differ in their structure, program of development and initiation of DNA replication. A library of approximately 5,000 mutants was constructed with a random insertion of transposon Tn5 in the genome of C. crescentus. The main objective of this work was to select possible mutants involved in DNA repair mechanisms which assist in the protection of this bacterium against the deleterious effects of ultraviolet light (UV) and methyl methanesulfonate (MMS). We have observed a total of 266 clones sensitive to one or both of these treatments. Close to 100 of the mutants were identified by nucleotide sequencing employing primers based on the transposon region. This technique allowed identifying not only the mutated gene by the transposon, as well as its relative position in the genome, that is eventual organizationin operons and possible SOS box in its neighborhood. The data generated made possible inferences about possible relationships and functions of those genes. The identification of genes which encode for conserved hypothetical proteins is interesting for the characterization of new genes related to DNA repair. The search of new bacterial models can enlarge our knowledge of the mechanisms of genome defense
