S-100 immunoreactivity is increased in white and gray matters of the contuse lesioned rat spinal cord (2005)

- Abstract: Objective: Astrocytes in the central nervous system produce S-100 protein, which is up-regulated after injury. It has been showing that the macromolecule helps to protect and to maintain neurons from neurodegeneration. Here, we analyzed S-100 expression in the contuse lesioned rat spinal cord promoted by the NYU Impactor. Methods and Results: Adult male Wistar rats (n=6) were submitted to a moderate degree of spinal cord contuse lesion. A 10g rod was dropped to the dorsal surface of the exposed spinal cord from distances of 25 mm. Another group of rats (n=4) were submitted to sham operation at T10-11 level. After 2 or 3 weeks, animals were sacrificed and their spinal cords sectioned, and processed for immunohistochemistry. S-100 immunoreactivity was detected by the avidin-biotin peroxidase technique and quantified by means of semiquantitative morphometric and microdensitometric image analysis. A strong S-100 reaction was observed bordering the lesioned site, in the region of the glial scar. Furthermore, S-100 immunoreactive cell profiles were also found to be elevated in the entire gray and white matters of the preserved tissue in cranial and caudal levels near the lesion. The S- 100 immunoreactive profiles were very similar to astrocytes showing a cell body and cell processes. S-100 reaction was seen in putative activated astrocytes. In the sham operated rats, S-100 reaction was detected only in the posterior funiculus close to the region of the laminectomy. Theresponses were seen 2 weeks after the lesion, being maintained by 3 weeks. Conclusion: The expression of S-100 in different regions of the lesioned spinal cord may be related to the formation of a glial scar close to the lesion and also to paracrine neurotrophic responses far from injury
