Basic fibroblast growth factor immunoreactivity after different degress of spinal cord contusion (2003)

- Abstract: Objetivo: Central nervous system induces a particularly rapid increase in the production of basic fibroblastic growth factor (bFGF) close to the site of lesion. This macromolecule helps to protect and to maintain neurons from secondary neurodegeneration. Here, we analyzed bFGF expression in the lesioned rat spinal cord following different degrees of local contusion. Métodos e Resultados: Adult male Wistar rats were submitted to different degrees of spinal cord contuse lesion. To promote the lesions, a 10g rod was dropped to the dorsal surface of the exposed spinal cord from distances of 12.5 mm, 25 mm or 50 mm, which characterize three groups: slight (SL), moderate (MD) or severe (SV) lesion, respectively. A group of rats was submitted to sham operation.After 90 days, animals were sacrificed and their spinal cords sectioned, and processed for immunohistochemistry. bFGF immunoreactivity was detected by the avidin-biotin peroxidase technique and quantified in anterior horn of cranial and caudal spinal cord sections close to the injury by means of semiquantitative morphometric image analysis. In cranial levels, the bFGF immunoreactivity was found to be elevated (p<0.01) following the MD (6%) and SV (5%) lesions compared to sham operated rats. Furthermore, no difference was observed in the cranial levels of the SL lesioned group compared to sham. In the caudal levels bFGF immunoreactivity increased (p<0.05) in the anterior horn ofthe rats that have received MD (7%) and SV (6%) lesion compared to sham rats; SL lesion did not trigger bFGF express. Conclusões: The results indicated that the expression of bFGF was increased in moderate and severe spinal cord contuse lesion at 90 days, which demonstrated that this neurotrophic factor may be involved in the local process against secondary neuronal degeneration as well as regenerative events
