Neuropeptide y, galanin and neurotensin systems in the nucleus tractus solitarii of adrenalectomized and kainate injected rats (2003)

- Abstract: Objetivo: The purpose of this study was to evaluate the effect of kainate stimulation on neuropeptide Y (NPY), galanin (GAL) and neurotensin (NT) immunoreactivities in the nucleus tractus solitarii (NTS) and their modulation by glucocorticoid hormones. Métodos e Resultados: Male Wistar Kyoto rats were submitted either to adrenalectomy (n=5) or sham-operation (n=10). Fourty-eight hours after surgery, the adrenalectomized rats received a single intraperitoneal dose of kainic acid (12mg/kg) (KAI/ADX). Furthermore, sham-operated animals were injected either with kainate (n=5) (KAI/SH) or vehicle (n=5) (SAL/SH). Eight hours after injections, rats were sacrificed and their brains processed for immunohistochemistry. Series of medulla oblongata sections were processed for NPY, GAL or NT immunoreactivity by means of indirect immunoperoxidase technique. Quantitative microdensitometric image analysis demonstrated a decrease in the area (mm2) occupied by NPY immunoreactivity in the NTS of KAI/SH (8721.8±790.1; p<0,05) as well as KAI/ADX (9846.3±521.8; p<0.05) both compared with SAL/SH (13650.8±937.5). Neurotensin immunoreactivity was decreased in KAI/ADX (339.1±36.15; p<0.05) compared with SAL/SH (433.6±65.8) rats although there was an increase in the KAI/SH (657.2±93.8; p<0.05) compared with SAL/SH. The intensity of galanin immunoreactivity (optical density) was decreased in the NTS of KAI/ADX (22.76±0.48; p<0.05) compared with SAL/SH (27.13±1.18) and KAI/SH(25.33±0.87; p<0.05). Conclusões: Kainate stimulations alters specific peptide immunoreactivities in the NTS which can be also modulated by glucocorticoids
