Hypothyroidism changes adenine nucleotide hydrolysis in synaptosomes from hippocampus and cerebral cortex of rats in different phases of development (2004)

- Abstract: Objetivo: The influences of thyroid hormones for the normal function of the mammalian central nervous system are dependent of the brain region and of developmental stage. Adenine nucleotides and their products also affect the CNS function, once that, ATP is an excitatory neurotransmitter, and adenosine has inhibitory effects on the neurotransmission. Then, we intended to study the effects of hypothyroidism on the hydrolysis of ATP to adenosine in hippocampal synaptosomes of rats in different phases of development. Métodos e Resultados: Hypothyroidism was induced in 60, and 420 day-old rats by thyroidectomy and methimazole (0.5%) added to their drinking water for 14 days. Neonatal hypothyroidism was induced by adding 0.02% methimazole in the drinking water from day 9 of gestation, and continually to at 14-day-old. Rats were killed and the structures dissected for the sinaptossomal preparation. The enzymatic activity was measured like described by Chan et al (Anal. Biochem., 157: 375-80, 1986), and all results where normalized with the protein content, which was measured by Bradford method (Anal. Biochem., 72: 218-254, 1976).Hypothyroidism increased the AMP hydrolysis in synaptosomes from hippocampus in 14-day-old (97%), 60-day-old (76%) and 420-day-old (33%) rats. Besides, ATP hydrolysis was increased by 17% in 420-day-old hypothyroid rats. Conclusões: Our results indicate an increase of the extracelullar adenosine levels in hippocampus inhipotireoidism, demonstrating the involvement of the 5'-nucleotidase in the synaptic function control in this pathology throughout brain development
