Genomic scale search for DNA repair genes in Caulobacter crescentus (2003)

- Abstract: The gram-negative bacterium Caulobacter crescentus, presents the anti-symmetrical division as an obligator characteristic of its cellular cycle. Two morphologically distint cells are produced in each division: a mobile cell and a fixed cell, also differing in relation to the development program and initiation of DNA replication. This explains why it is considered one of the main models of study for cellular differentiation in bacteria, whose molecular mechanisms come being detailed in the last years. A library of approximately 5,000 mutants was constructed with a random insertion of transposon Tn5 in the genome of Caulobacter crescentus. The main objective of this work was to select possible mutants involved in DNA repair mechanisms, that assist in the protection of this bacteria the ultraviolet light (UV) and methyl metanesulfonate (MMS). A screening was carried through by the search of sensitive clones to these genotoxic agents. The screening was performed in genomic scale (96 clones/test) and the sensitive candidates were confirmed at least three times. Among 5,000 initial clones, we observed that 156 were sensitive to UV-light and 112 to MMS and 29 to both treatments. These mutant clone candidates are being used to identify the genes interrupted by Tn5, through DNA sequencing. We hope this genomic strategy will allow us to have the profile of genes involved in repair of DNA damage for Caulobacter crescentus
