Connexin 31.1 is expressed in mouse retina and modulated by dark adaptation (2003)

- Abstract: Purpose: The channels formed by connexins (Cx) allow the passage of small molecules and ions, which in excitable cells provide cell communication by means of eletrotonic potential. Different connexins have been detected in distinct tissues, and some of them were detected by RT-PCR in mice retina. In retina, conductance of these channels is decreased by neuromodulators, such as nitric oxide and dopamine, which in turn are related to ambient light levels. Although these neuromodulators are known to uncouple cells, their effects on connexin gene expression are still poorly understood. Our aim was to determine the expression of specific connexins (Cx31.1, Cx36 and Cx45) in the mouse retina, and their possible modulation by dark adaptation. Methods and Results: 24 adult mice divided into 3 groups (Control, 1 day and 7 days dark-adapted) were sacrificed, and their retinas were dissected for RNA isolation by using TRIzolÒ. mRNA was reversed transcribed and cDNA was amplified with a 5700 SDS Real-Time PCR machine.Data were entered into a multivariate analysis of variance (MANOVA), followed by pairwise comparisons (Tukey's HSD test). Cx31.1 showed a significant increase (P<0.05) while Cx36 decreased (P<0.01) after 7 days of dark. Cx45 gene expression decreased for 1 day of dark adaptation (P<0.05). Conclusion: Some connexins were not detected in previous studies using RT-PCR, and consequently were not further analyzed by othertechniques, such as immunoblot and immunohistochemistry. Using a more precise technique such as real-time PCR we were able to detect Cx31.1 transcripts in mice retinas and state that its expression is upregulated by dark adaptation. Although not statistically significant, a discrete increase of Cx31.1 expression was also found for 1 day dark-adapted retinas, indicating that probably the modulation of this connexin may start as early as 1 day of dark adaptation
