Effects of spontaneous running on glial and neurochemical plasticity in rat brain (2003)

- Abstract: Objetivo: Physical activity triggers the expression of neurotrophins in the hipocampal formation (HF) and other brain areas which may favor local plasticity, a necessary molecular event for learning, memory and general cognitive functions. This work analysed the effects of spontaneous activity in the running wheels in the state of astroglial and microglial activation as well as in the content of nitric oxide synthase (NO) immunoreactivities (IRs). Métodos e Resultados: Young male rats were placed in individual cages with running wheels to allow spontaneous activity (running wheel group RW, n=16) during 4 or14 days. Control rats were placed in individual cages without wheels (sedentary group SED, n=16). Immunohistochemistry was employed and quantified by means of morphometric image analysis (KS400, Zeiss). The distance (m)/12h was: about 600 in the 4 day and 400 in the 14 day groups. The VO2 (ml/kg) was: about 6000 in the 4 day and in the 14 day groups. The VCO2 was: about 5000 in the 4 day and in the 14 day groups.) The area of the GFAP IR increased (p<0.05, indication of astroglial reaction) in the CA1, CA2, dentate gyrus (DG) and stratus lacunosun (SL) of the HF of the 14 day RW rats but not in the frontal region of the cerebral cortex (FroCo) and nucleus caudate putamen (CP) of these animals. Furthermore, GFAP IR did not change in the above studied regions of the 4 day RW rats. The area of the OX42 IR increased (p<0.05, indication of microglial reaction) in theCA1, CA2, DG, SL, FroCo and CP of the 14 day trained rats. OX42 IR was also found to be elevated (p<0.05) in the CA2, DG, SL and CP, but not in the FroCo of 4 day RW rats. NOS immunoreactive (ir) neurons were observed in the subregions of the HF, CP, neocortex and Acc. Many NOS ir terminals were seen in the Acc. Quantitative analysis demonstrated a decreased number of NOS ir neurons in the CP and Acc of 4 days RW trained rats. Conclusões: The results indicated that RW running interferes with the glial and neurochemical plasticity in rat brain
