- Abstract: Polymersomes, vesicles formed by amphiphilic copolymers, are more structured than lipid vesicles and can be used as nanoreactors, aggregates that can sequester reagents or enzymes in the internal compartment, shielding them from external agents and facilitating their transport and applications in the most diverse environments. OBJECTIVES: Obtaining stimuli-responsive amphiphilic copolymers capable of forming polymersomes. MATERIALS AND METHODS: Poly(dimethylaminoethylmethacrylateblock-polystyrene), PDMAEMA-b-Sty, copolymers were synthesized using RAFT (Reversible Addition-Fragmentation Chain Transfer). The copolymers were characterized by SEC-IR, FTIR and 1H/NMR. Hydrodynamic diameters were measured by DLS. The presence of a hydrophilic compartment in the polymersomes was estimated by EPR, as follows: polymersomes were prepared in a solution containing a spin label followed by the addition of ascorbic acid that suppress probe’s signal outside the vesicle. Residual EPR signal was assigned to the existence of an internal aqueous compartment. RESULTS AND DISCUSSION: Chemically characterized (NMR, FTIR, SEC-IR) amphiphilic block copolymers were obtained. Polymers responded to pH and temperature by changing their aggregation pattern. Under simple compatible solvent dissolution, the polymers assembled into aggregates with dimensions that match with the formation of vesicles (polymersomes). Entrapment of EPR probes indicated that the polymersomes possessed internal aqueous compartments of 0.59 and 0.53 % of sample’s total volume (1mg of copolymer in 1mL solution), respectively for PDMAEMA315-b-Sty244 and PDMAEMA315-b-Sty781. CONCLUSION: Stimuli-responsive amphiphilic copolymers capable of assembling into polymersomes that can be used as nanoreactors or drug delivery systems were synthesized and characterized.
