Effect of inoculum preparation of Kluyveromyces marxianus in the hemicellulosic hydrolysate on cellulosic hydrolysate fermentation (2019)

- Abstract: In this work, rice straw hemicellulosic or cellulosic hydrolysates were evaluated as inoculum media for cellulosic ethanol production by thermotolerant yeast Kluyveromyces marxianus NRRL Y6860. The rice straw was pretreated with mild alkaline solution followed by dilute acid pretreatment to attain the hemicellulosic hydrolysate. The solid recovered (cellulign) was submitted to an enzymatic hydrolysis in vertical ball mill reactor with high solids content (20% w/v). The cellulosic hydrolysate containing 107 g/L glucose was supplemented with nutrients and used as fermentation media. The results showed that K. marxianus was able to growth in both hydrolysates (hemicellulosic or cellulosic) containing 20 g/L of xylose or glucose, respectively. However, the fermentative performance of K. marxianus was strongly impacted by the hydrolysate type used for inoculum preparation. The highest fermentation parameters (YP/S of 0.45 g/g and QP of 3.91 g/L.h) were obtained when the inoculum was prepared in glucose (cellulosic hydrolysate). These values were, respectively, about of 1.2 and 2.8 fold in relation to the inoculum prepared in xylose (hemicellulosic hydrolysate). These results reveals that the use of hemicellulosic hydrolysate for cell growth affects the yeast fermentative performance and, therefore it is not adequate to prepare the inoculum cells aiming to cellulosic ethanol production.
