- Abstract: Objectives: Owls possessa sophisticated visual and auditory system. Visual information is provided through two main pathways: the tectofugal and the thalamofugal pathways. Thereis only limited information about the neuroanatomy/chemistryof the visual Wulst and other visual and auditory centers in the telencephalonandthalamus,thus we investigatedthese areas byhistological, immunohistochemical and retrograde tracing methods in thebarn owl. Methods: FiveBarn owls were perfusedandthe brainswere cut on a freezing microtome. Standard histological staining methods such as Nissl-,Gallyas and cytochrome oxidase staining as well as immunohistochemical techniques using antibodies to NeuN, parvalbumin, calbindin, and DARPP-32 were performed. In one owl, the retrograde tracer FluoroGold was injected in the Wulst. Results: The different layers of the Wulst could be best outlined by DARPP-32 and cytochrome oxidase staining. Primary sensory areas like the relatively small-sized visual entopallium and the large auditory Field L2 were characterized by the absence of DARPP-32.Parvalbumin staining outlined a relatively small nucleus rotundus and a huge OPT, the relay nuclei of the tectofugal and thalamofugal system, respectively, as well as the auditory nucleus ovoidalis. Retrograde tracing experiments together with NeuN staining indicated that the OPT consists of several subnuclei. Conclusions: Results indicate that the thalamofugal system of owls is moreprominently developed than the tectofugal system, which is reflected by the respective size of their thalamic relay nuclei as well as their telencephalic targets. The large size of the auditory Field L and its thalamic relay nucleus ovoidalis should motivate further studies to investigate the role of these areas in auditory processing.
