- Abstract: Introduction and Objectives : Plasmodium infection results in activation of T, NKT and NK lymphocytes, which has been implied in the initial control of parasitemia, through IFN- ã - mediated effector mechanisms. NKT cells can be distinguished by the expression of NK cell markers such as CD161 (NK1.1), a molecule involved in cell activation, IFN-ã production and cytotoxicity. However, NKT cells represent a heterogeneous population since not all CD1drestricted T cells express NK1.1 and some NK1.1+ T cells are not restricted by CD1d. Moreover, conventional T cells increase NK1.1 expression when activated, whereas a decrease is observed in CD1d-restricted T cells. Thus, this study aims to characterize NK1.1+ T cells in the acute response to P. chabaudi. Methods and Results : M ultiparametric flow cytometric analysis of splenocytes obtained from C57BL/6 mice on days 0, 4 and 7 of infection with 106 P. chabaudi parasitized erythrocytes revealed an increase of CD3+NK1.1+ cell frequency at the different time points, being the total number of these cells higher on day 7.Among CD3+NK1.1+ cells, CD4+, CD8+ and CD4-CD8- cells represented 35,83%, 21,38% and 42,79%, respectively. Numbers similar to those were found in infected CD1dKO mice. CD4+NK1.1+ and CD8+NK1.1+ cells showed an activated phenotype (CD69 high, CD45RBhigh, CD44high, and CD62Lhigh) and produced IFN-ã. A higher expression of Fas and FasL was detected in CD8+NK1.1+ cells at day 7 ofinfection. Analysis of CFSE-labeled cells kept in culture for 72h showed lower spontaneous proliferation of NK1.1+ cells compared to NK1.1- cells, for both CD4+ and CD8+ lymphocytes. When stimulated with plate-bound anti-CD3 antibodies, a strong proliferative response occurred at day 0, decreasing after infection with NK1.1+ cells showing the lowest scores. Conclusion: These results suggest that CD4+NK1.1+ and CD8+NK1.1+ cells have effector functions that may contribute to the acute immune response to P. chabaudi.
