In vitro intra-pleural response of chronic Trypanosoma cruzi-infected mice to challenge with parasite antigen (2006)

- Abstract: INTRODUCTION AND OBJECTIVES: CD8+ lymphocytes are involved in T. cruzi control but their in vivo dynamics are poorly understood. To elucidate the involvement of the different CD8+ cell subsets in the tissue response to the parasite, we analyzed the local population changes in chronically-infected mice after intrapleural challenge with T. cruzi antigen. METHODS: Chronic mice were challenged intrapleurally with T. cruzi antigen (equivalent to 108 culture trypomastigotes), the effects on the pleural exudate being estimated by FACS, 24-48 hours later, in relation to unchallenged chronic mice. RESULTS: In vivo challenge with T. cruzi antigen resulted in increased number of pleural exudate cells. Polymorphonuclear cells (PMN, Ly6GHIGH) dramatically increased in the first 24 h (25 fold), but their numbers slightly declined at 48 h (16 fold increase in relation to control). Macrophages (Mac1HIGH, Ly6GINT) were already present in the pleura of chronic unchallenged mice; after challenge, they steadily increased at 24 h (2,5 fold) and 48 h (6,7 fold), becoming the predominant population at this late time point.While B cells (B220+) were not increased in the pleural exudate following challenge, CD8+ and CD4+ T cells showed an important increase at 24-48 hours, the number of CD8+ cells exceeding those of CD4+ cells. Thus, while the CD4/CD8 cell ratio in the pleural exudate of chronic unchallenged mice was around 1 (similar to that on theblood), 48 hours after challenge it dropped to 0,3-0,5. Analysis of CD8+ subsets in the pleural exudate revealed that CD8HIGHCD45RCLOW (CD8HL) and CD8LOWCD45RCLOW (CD8LL) cells showed the highest increase. The percentage of pleural exudate CD8+ cells expressing CD69 was already high in unchallenged chronic mice, moderate or no further increases being observed after antigen challenge. Among CD8+ subsets, CD8HL cells displayed the highest frequency of CD69+ cells. CONCLUSIONS: Intra-pleural stimulation of chronic mice with T. cruzi antigen results in a copious inflammatory infiltrate dominated at 24 h by PMN cells, and by macrophages at 48 h. Among lymphocytes, an inverted CD4/CD8 ratio was observed, with predominance of CD8HL and CD8LL cells.
