- Abstract: Introduction and objective: During the chronic phase of Experimental Chagas Disease, challenge with trypomastigotes results in activation of the immune system with consistent reduction of the subpatent parasitemia levels. Due to the high numbers of mononuclear phagocytic cells, the liver plays a major role in the clearance of blood trypomastigotes. In this work we investigated the changes in intra-hepatic leucocytes of chronic infected mice following challenge with trypomastigotes. Methods: Experiments were carried out in 10 month-infected chronic C57BL/6 mice, 24 and 48 hours after challenge with 5 x106 trypomastigotes. Intrahepatic leukocytes were isolated by Metrizamide density gradients after treatment of the hepatic tissue with collagenase. Results: We verified that 24 hours after challenge the numbers of infiltrating cells in the hepatic tissue were much higher than in chronic mice, the infiltrating cells being mainly distributed around the vessels. Phenotypic characterization of the infiltrating cells revealed that the most expanded population in the liver of challenged mice was that of Mac1+ cells, followed by NK cells (NK1.1+CD3-), the number of CD4+ and CD8+ cells being also notably augmented.Challenged mice CD4+ and CD8+ liver cells showed increased CD69+ cell frequencies compared to chronic control mice. Besides this, a notable augmentation in the frequency of D25+CD122+ cells was observed among CD4+ cells 24hafter challenge, that was followed at 48hs by an increase of CD4+CD25-CD122+ cells. After 24h hours of challenge, intrahepatical CD4+ T cells displayed a notable increase of IFN-ã spontaneously-producing cells, these numbers showing no further increases after in vitro stimulation with anti-CD3/CD28 antibodies. In contrast to this, intrahepatical CD8+ T cells only produced this cytokine after in vitro stimulation. Conclusions: We conclude that challenge of chronically-infected mice with trypomastigotes rapidly increases the T. cruzi specific response, the liver displaying an important infiltration by Mac1+ cells and activated lymphocytes that complement the important role played by this organ in parasite clearance.
