Ikaros expression by B-1 cells (2006)

- Abstract: INTRODUCTION: The hematopoietic system in mammals is continuously regenerated by a rare population of self-renewing, multipotent stem cells resident within the adult bone marrow. Transcription factors regulate gene expression by inducing activation of lineage specific genes or silencing the unspecific ones, driving in this way the commitment of cell with a definite lineage. In the hierarchal model the hemapoietic stem cells branch early in myeloid and lymphoid progenitors. The transcription factors involved in B-lymphocytes development are well established. Ikaros, a DNA binding protein, is pointed as central in this cell lineage fate. We have shown that B-1 cells spontaneously express both myeloid and lymphoid restricted transcription factors. They can differentiate in phagocytes losing of lymphoid genes expression and retaining a network of transcription factors related to myeloid lineage. The promiscuous expression of myeloid and lymphoid transcription factor networks in a differentiated cell challenges the on/off paradigm of hematopoietic cell differentiation.Taking this investigation further, here we searched for the possible expression of Ikaros by B-1 cells. METHODS AND RESULTS: Ikaros expression was analyzed by RT-PCR (mRNA) and western blotting (protein). B-1 cells were obtained from detached cells after culture of peritoneal fluids and the phagocytes were derived from B-1 cells after re-attachment as described inPopi et al (submitted). We found that both B-1 cells and their derived phagocytes express different isoforms of Ikaros, probably involved in the different expression patterns of these cells. CONCLUSION: Despite the promiscuous lineage transcription factors expression by B-1 cells; we have shown that they also express Ikaros, which are known as chromatin remodeling factors in lymphoid cell commitment. The characterization of Ikaros isoforms, in B-1 cells will be helpful to understand the hematopoietic cell differentiation.
