Skeletal muscle damage induced by cryolesion is attenuated in the induced 70-KD heat shock protein (HSP70i) transgenic mouse (2005)

- Abstract: Objetivo: It is well known that certain hsps have been for long recognized as tissue protectors, however, their role on skeletal muscle damage and regeneration is still unclear. This work examined the role of the hsp70i in cryolesioned skeletal muscles. Métodos e Resultados: : Transgenic mice overexpressing hsp70i and age-matched controls were submitted to cryolesion of soleus and tibialis anterior (TA) muscles and were evaluated at 1, 10 and 21 days after lesion. Another group of control mice was treated with radicicol (0.1 mg/20 g of body weight), an inductor of heat shock response, immediately after cryolesion. The muscles were stained with toluidine blue and ac-phosphatase and immunostained with antibodies against N-CAM, Ki-67 and MHCd/n. The results clearly show that hsp70i mice and radicicol treated mice had reduced incidence of necrosed myofibers in soleus (41.4% and 37%, respectively) and TA (53.5% and 55%, respectively) muscles when compared to control mice at 1day post lesion. The percentages of N-CAM-positive satellite cells (SCs) were increased at 1 and 10 days post lesion in soleus (2.6 and 3.9 fold, respectively) and TA (6.9 and 3.9 fold, respectively) muscles only in control mice. The ratio between the fiber cross-section area and body weight (CSA/bw; um2/g) was reduced at 10 days after cryolesion only in soleus (2.6 fold) and TA (1.6 fold) muscles from control mice. HSP70i transgenic mice had neither cryolesion-induced reduction in fiber CSA norincreased N-CAM positive SCs. Finally, the expression of Ki-67-positive proliferating cells and MHCd/npositive differentiating cells at 10 days were decreased in Hsp70i mice when compared to that of control mice. Conclusões: These results suggest that hsp70i attenuates skeletal muscle damage induced by cryolesion
