Expression of a treptococcal M protein epitope fused at different sites of flagellin (FliCd) as an approach for the development of bivalent vaccines based on attenuated Salmonella strains (2003)

- Abstract: Flagellin, the flagellar filament monomer, has been used as a carrier for heterologous epitopes genetically inserted at the central hypervariable region (region IV). Recombinant flagellins are often functional and can elicit humoral responses to both flagellin and heterologous epitope in mice following parenteral immunization. In the present work we investigated the immunogenicity and vaccine potential of attenuated Salmonella strains able to express recombinant flagellins genetically fused at different sites to a 15 mer linear epitope of Streptococcus pyogenes M5 protein as an approach to identify alternative sites for the integration of heterologous epitopes at the FliCd flagellin. The peptide was inserted at flagellin regions presenting reduced (region IV), partial (region III and VI) or complete homology (region VIII) among several fliC alleles. All regions, with exception of the highly conserved region VIII, accepted the insertions without loss of motility, albeit, in some cases, motility was significantly reduced during in vitro growth. Only mice immunized parenterally with live Salmonella strains expressing heterologous flagellin fused with the M5 protein epitope at region IV and VIII were fully protected after challenge with S. pyogenes. The same recombinant flagellins with fusions at regions IV and VIII were also able to elicit serum antibody responses to the heterologous peptide as determined in ELISA assays. In contrast to fusions at other flagellinintegration sites, Salmonella strains expressing recombinant flagellins with the insert at region VIII produced soluble molecules unable to assemble into morphological normal flagella. Based on the present evidences we suggest that regions other regions besides region IV can also be used for the genetic fusion of heterologous epitopes at the Salmonella flagellin as an approach to engender peptide-based bivalent vaccines. These results further demonstrated that expression of epitopes derived from the M5 protein genetically fused to Salmonella flagellin represents a promising alternative for the development of anti-streptococcus vaccines without the risk to elicit autoimmune responses
