Occurrence and methods to identify Legionella spp and legionella-like amoebal pathogens in environmental samples and man-made systems (2003)

- Abstract: The aims of this research were to verify the occurrence of Legionella spp and Legionella-like Amoebal Pathogens on the environmental samples and in man-made systems; to evaluate the most sensible method to detect these bacteria in environmental samples and to characterize the isolated strains of Legionella spp. Water samples were collected from four different environments: water reservoirs; dental units; cooling towers and boilers. The samples were concentrated by membrane filtration, treated by heat and submitted to isolation, direct imunofluorescence antibody and coculture using amoebae. Concentrated samples were plated on a selective aBCYE agar; submitted to direct fluorescent antibody staining using polyclonal antibodies specific to L. pneumophila serogroups and to other 21 specimens of Legionella, and to amoebic coculture procedure using an axenic culture of Acanthamoeba castellanii. The best results for detection of Legionella were culture on aBCYE. In the environments sampled up to this moment, Legionella spp was isolated from water and biofilm samples from cooling towers. The results obtained showed that L. pneumophila serogroup 1 was the only species present in the samples tested. Coculture procedure of sample with A. castellanii improved the culturability of Legionella spp on aBCYE but more specific tests in vitro are necessary to study these interaction
