Antibody inducing properties of a prototype bivalent herpes simplex virus/enterotoxigenic Escherichia coli DNA vaccine (2003)

- Abstract: The immunogenicity of a viral/bacterial bivalent DNA vaccine (pRECFA), expressing a hybrid protein containing N (amino acids 1 to 27) and C (amino acids 269 to 394) terminal sequences of the Herpes Simplex Virus type 1 (HSV-1) glycoprotein D (gD) genetically fused to the complete amino acid sequence (147 amino acids) of the major structural subunit of enterotoxigenic Escherichia coli (ETEC) CFA/I fimbriae (CFA/I), was evaluated following intramuscular (i.m.) administration in BALB/c mice. The protein was in vitro expressed as a surface-exposed membrane protein in pRECFA-transfected BHK-21 cells. The hybrid protein proved to be immunogenic in vivo and significant serum antibody levels against CFA/I or HSV were detected in all or half of the pRECFA-vaccinated, respectively. Serum antibody response induced in mice immunized with the DNA vaccine belonged predominantly to the IgG2a subclass and detected the respective bacterial and viral antigens, either intact or heat-denatured.Agglutination of CFA/I+ ETEC as well as binding properties of the native fimbriae was not inhibited by pRECFA-induced antibodies. On the other hand, gD specific antibodies preserved virus neutralization activity as observed in mice immunized with DNA vaccine encoding the full length protein. These results suggest that the present HSV-ETEC DNA vaccine represents a promising approach for the activation of antibody responses targeting continuos epitopes of antigensinvolved with pathogenicity, as surface-exposed adhesins and secreted toxins, of viruses and bacteria
