- Abstract: It has been shown that inflammatory stimulus in the brain increase excitatory glutamatergic transmission especially at the level of NMDA receptors. These receptors have an important role in glutamate neurotoxicity and are in general coupled with the generation of NO through the activation of nNOS. Our aim was to test whether NMDA-NO pathway is involved in LPS activation of NF-kB in different areas of the brain. Cerebellum, hypothalamus, hippocampus, pre-frontal cortex, and striatum were isolated from male Wistar rats treated previously for 2-24 h with LPS (1mg/kg, i.v.) or saline. Gel shift assays showed that LPS activated NF-kB in all structures tested. Supershift assays indicated that the major DNA-protein complex altered by LPS corresponded to p50/p65 heterodimer. MK-801(0.3-3 mg/kg) or 7-Ni (5-25 mg/Kg), injected i.p. 20 min before LPS, partially reduced LPS effects. This reduction were not further potentiated in rats treated with both compounds, suggesting that glutamate acts on NMDA-nNOS pathway to activate NF-kB in response to LPS
