- Abstract: Objetivo: We showed previously that activation of oxytocinergic (OT) projections from hypothalamus (HYP) to the nucleus tractus solitarii (NTS) facilitates reflex bradycardia and impairs exercise tachycardia in trained rats (AJP2000;278:R1474 and AJP 2002;282:R537). We sought now to identify the effects of hypertension and training (T) on mRNA expression of OT in projection and biosynthetic areas. Métodos e Resultados: Male SHR and WKY were submitted to T (treadmill, 50-60% VO2 peak for 13 weeks) or kept sedentary (S); tail pressure was measured weekly. Brains were removed, frozen and sliced to isolate dorsal brainstem (DBS) and HYP areas; total RNAs were extracted and used for RT-PCR amplification.T-induced pressure fall was observed only in SHR group.Brain OT mRNA content was differentially affected by hypertension (HYP -39%; DBS +108% AU, SHRS vs WKYS) but significantly increased by T in both areas of either group, with the largest increment (3.5-fold) being observed in DBS. In situ hybridization for OT35S-oligonucleotide in brainstem slices of WKYT and WKYS confirmed similar T-induced changes (3.5-fold) restricted to the NTS. Conclusões: Specific training-induced increase in OT mRNA expression in the NTS, by slowing down the heart might explain the reduced exercise tachycardia of SHRT and WKYT. Opposite plastic changes in brain OT circuitry after hypertension and T reinforce the efficacy of T to correct hypertension-induced changes.
