- Abstract: To validate a new technique for digital evaluation of gingival inflammation using intraoral scan (IOS) files. 57 participants diagnosed with moderate gingivitis were randomized to follow Mechanical plaque control/chemical plaque control using Multi-Component Oral Hygiene Regimens Dual Zinc plus arginine and CPC mouthwash for 9 months. Gingival index and plaque index scores obtained, and a (3Shape-TRIOS 3) scanner used to capture IOS at the same clinical visit. Quality inspection software (Geomagic Controlx) was used to measure digital variables. After tracing the gingival margin, The region of interest (ROI) for gingival margin color (GMC) was determined by a 2mm offset from tooth-gingival contact (TGC) on gingival direction. ROI for TGC Curvature was determined at 0.5mm offset on both directions at the level of gingival margin.Clinical and digital variables of 110 scans were evaluated. The regression analysis revealed a significant relationship between gingivitis and CILAB color coordinate B* (Blue-Yellow), A* (Red-Green) (p < 0.001) also HSV color saturation revealed significant linear relationship, Increased color saturation associated with increased severity of gingivitis. Plaque index exhibited a significant linear association with the TGC Curvature, both in the absence and presence of disease (p = 0.0001). Digital analysis of GMC and TGC presents a precise and highly reproducible technique for the detection of gingival inflammation. This method can be a valuable tool for dentists and researchers in the diagnosis and/or monitoring of gingivitis
