Repair of calvaria critical defects in rats subjected to Epimedium Sagittatum extract standardized on icariin (2014)

- Abstract: Various reports have identified icariin as a promising osteogenic agent. We sought to evaluate the influence of Epimedium Sagittatum extract standardized on icariin on bone neoformation of critical-sized defects in rat calvaria. An 8-mm diameter bone defect was created in each of 40 rat calvaria with a trephine drill under continual saline irrigation. Animals were randomly divided into a test group receiving Epimedium Sagittatum extract of icariin (5,8mg/mL) and a control group receiving equal volume of saline solution. Both substances were administered through a feeding tube until the day of euthanasia. After each observation period of 7, 14, 21, and 42 days, five animals from each group were euthanized. Calvaria defects samples were fixed in 10% formalin until their subsequent x-ray and histological processing and evaluations. The evolution of repair analyzed reduction of bone defect area on the x-ray image and histomorphometric amount of bone formation between the groups. X-ray images showed that the defect area in the test group was significantly smaller than in the control in allobservational periods. Histomorphology revealed significant osteogenesis in the test group at 42 days and histomorphometric analysis showed significant osteogenesis in the test group at 7 days (P = 0.021), 14 days (P = 0.014), 21 days (P = 0.021), and 42 days (P = 0.009)
