The role of glucocorticoids in the glycerol-3 phosphate generation pathways in epididymal and retroperitoneal adipose tissue of rats (2013)

  • Abstract: Introdução The glucocorticoids are important hormones involved in the control of lipid, carbohydrate and protein metabolism in mammals. It is often associated with obesity and show opposite effects on lipid metabolism depending on the localization of the adipose tissue. The biochemical mechanisms involved in these effects are not yet totally elucidated. Objetivos The objectives of the present study are to determine the effects of dexamethasone chronical treatment on the glycerol-3-P (G3P) generation pathways for the triacylglycerol synthesis in two different white adipose tissue of rats: epididymal and retroperitoneal. Métodos For this, male Wistar rats (150-200g) were treated intraperitoneally with dexamethasone phosphate (DEX, 1mg.Kg-1 for 7 days) or saline during the same of time (CON). The animals were killed by cervical dislocation 24 hours after the last injection and the epididymal (EPI) and retroperitoneal (RETRO) adipose tissues were removed. This study was approved by the Ethics Committee on Animal Experimentation of the Faculty of Medicine of Ribeirão Preto - USP (number 187/2010).Resultados We observed that DEX treated animals showed an increase in weight (~30%) of both EPI and RETRO WAT, which was also associated with an increase in the total lipid content (~20%). In EPI WAT the DEX treatment caused: a marked increase in the rate of glucose uptake (in vitro), an increase in the glycerokinase (GyK) activity and a decrease in glyceroneogenesis, estimated by the 1-14C-pyruvate incorporation into total lipids and by phosphoenolpyruvate carboxykinase (PEPCK) activity (~30%). On the other hand in RETRO adipose tissue, DEX caused an increase in glyceroneogenesis estimated by the increase in 1-14C-pyruvate incorporation in glycerol-TAG and a decrease in GyK. In the epididymal WAT there is a clear reciprocal effect of DEX between the glycolysis activity and glyceroneogenesis for G3P formation, what was not observed in retroperitoneal WAT. Conclusão These data suggest that glucocorticoids play an important role in modulating G3P generation pathways in WAT for TAG synthesis, showing a clear specific effect depending on the body localization of the white adipose tissue.
