Uroguanylin inhibits the (Na++K+)ATPase activity in LLC-PK1 proximal tubule cells (2013)

- Abstract: Introdução Uroguanylin (UGN) is a small guanylyl cyclase-activating natriuretic peptide produced in the kidney in response to hypernatremia with autocrine/paracrine functions including modulation of renal sodium excretion. Usually, the effects of this peptide on renal sodium balance are associated to changes in proximal tubule (PT) sodium reabsorption. However, the mechanisms involved in this process are not completely known. Objetivos In the present work, we tested the possibility that the modulation of the (Na++K+)ATPase by UGN could mediate the effect of this peptide on the PT sodium reabsorption.Métodos LLC-PK1 cells, a porcine proximal tubule cell line, were maintained in DMEM supplemented with 10% fetal bovine serum at 5% CO 2 /37 o C, for 48 or 72 h. After confluence, cells were washed and deprived from serum overnight (starvation). During starvation, cells received or not treatment with UGN (10 -12 to 10 -6 M). The (Na + +K + )ATPase activity was measured as described previously (J. Biol. Chem. 256:3718-3727, 1981). Immunoblotting assay was used to determine enzyme expression. Resultados UGN inhibited enzyme activity in a dose-dependent manner with effect of 51.89±3.4% inhibition at 10 -8 M (n=3). This effect was reversed by 10 -6 M KT5823, a selective inhibitor of PKG (n=2), but not by 10 -6 M U0126, a potent specific inhibitor of ERK1/2 (n=3). Moreover, UGN effects on the sodium pump was mimicked by 10 -6 M cGMP (n=3). Furthermore, it was observed that 10 -8 M UGN decreased in 37.97±5.28% (n=3) the expression of (Na + +K + )ATPase α1 subunit. In addition, 10 -6 M cGMP or 10 -8 M UGN inhibited (32.0±3.8%; 45.87±5.5%, respectively) PKB expression. Conclusão The present study indicate that UGN inhibits the proximal tubule (Na + +K + )ATPase activity and expression and this effect involves cGMP.
