Cytoprotective activity of leaf crude extract of eugenia uniflora and quercetin in insulin-producing cells. (2010)

- Abstract: OBJETIVOS: Diabetes mellitus is induced, at least partially, by oxidative stress. Eugenia uniflora L. leaves are used in popular medicine as alternative to the treatment of this disease because they are rich in phenolic compounds which have many protective/preventive properties. The aim of this paper was to evaluate the cytoprotective activity of leaf crude extracts and a bioactive compound commonly found in these extracts: quercetin. MÉTODO E RESULTADOS: Acute and chronic activity of different concentrations of the extracts and quercetin was evaluated morphologically (HE stain) and by MTT method. Acute activity was performed through pre-incubation of these substances with RINm5f cells for 1h and posterior induction of oxidative stress by hydrogen peroxide (150μm/2h). On the other hand, chronic exposure was performed through 24h incubation with the replacement of the medium before addtition of hydrogen peroxide solution. Acute essay showed a reduction in the viability of the control group of nearly 33%. Cytoprotection was partially achieved with aqueous extract (50μg/ml) and quercetin. As to quercetin, the result was dose-dependent (6.25 > 3.12μg/ml). Chronic essay showed a reduction in the viability of the control group of nearly 45%.The cytoprotection achieved with aqueous extract (75 and 50μg/ml) was about 16.5% and with alcoholic extract (250μg/ml) nearly 18%. The cytoprotection achieved with quercetin was 12% (6. 25μg/ml) and 25% (3.13μg/ml). Through HE stain was verified a loss of intercellular adhesion and membrane/shape cell alterations. Aqueous extract (50μg/ml) showed partial prevention in acute essay with a slight increase in cellular prolongations and a decrease of membrane alterations. There was a more intense prevention of damage by 3.13μg/ml quercetin (cells with morphology similar to the control group). At chronic essay, aqueous extract and quercetin (75 and 3.13μg/ml, respectively) partially prevented cell damage. There were still some round cells but cell prolongations were preserved. Cells with quercetin concentration of 6.25μg/ml showed morphology similar to control group with few round cells. CONCLUSÃO: These effects suggest that the tested compounds reduce oxidative stress, probably by their direct (acute essay) and indirect (chronic essay) antioxidant activity.
