- Abstract: Introduction: The mammalian pineal gland presents a circadian rhythm of melatonin synthesis and secretion that is imposed by noradrenaline stimulation at night. Cocaine, which is an inhibitor of noradrenaline recapture, promotes increased synthesis of melatonin in explants of pineal gland. An important metabolite of cocaine, when used in the form of "crack", is the AEME (methylecgonidine), which has effects on the central nervous system even more intense than cocaine. Objectives: The aim of this work was to study the actions of AEME on melatonin synthesis, investigating its effects in vitro, in primary cultures of pineal glands, and in vivo, analyzing the daily profile of melatonin. Methods: Pineal glands were isolated from Wistar rats and maintained in culture for 48h, in BGJb medium. Then, they were stimulated by noradrenaline (1µM) in association with AEME in different concentrations (1µM and 100µM), that was added 30 min or 48h before noradrenaline addition. Melatonin synthesis was evaluated by HPLC with electrochemical detection. Results and conclusions: AEME reduced melatonin synthesis in every concentration used, independently of incubation time (Nor 1µM = 10,56±0,61ng/gland; Nor + AEME 1µM = 7,56±0,69ng/glnd; Nor + AEME 100µM = 7,96±0,78ng/gland; Nor + AEME 100µM (48h) = 7,93±0,72ng/gland). Thus, AEME seems to inhibit melatonin synthesis in the rat pineal gland, by a yet undetermined mechanism.
