Molecular typing of histoplasma capsulatum isolated from bats captured in São Paulo State, Brazil (2006)

- Abstract: Little is known about the genetic strain diversity and geographical range of the Histoplasma capsulatum isolated from bats in Brazil. This has prompted us to characterize 15 H. capsulatum isolated from liver and spleen from 963 captured bats in the urban areas of the São Paulo State. The fungi were identified by its phenotypic characteristics. The genotypic relatedness was carried on using a PCR fingerprinting assay performed using primers (GACA)4 and M 13 and generated amplicons with considerable polymorphism. Cluster analysis of the generated DNA banding patterns with primer M13 revealed two groups with 39% similarity. Group I included 6 H. capsulatum isolates, and Group II was composed by 9 samples. DNA fingerprinting with primer (GACA)4 was less discriminatory than M13. A major cluster composed by 12 H. capsulatum isolates was generated exhibiting from 40to 100% genetic relatedness. H. capsulatum 523, 154, and 189 isolates showed 100% similarity with both primers, suggesting that an environmental microniche could be acting as sources of infection for animals and the local human population. This is a preliminary study; however, the high genetic polymorphism observed also suggests a different H. capsulatum population in São Paulo State
