Interaction of signaling molecules and PKCS in phagocytosis via FC G receptors mediated by leukotrienes (2006)

- Abstract: Introduction and Objectives: It is been shown that LTB4 and cysLT enhances alveolar macrophages (AM) FcR-mediated phagocytosis dependent on protein kinase C (PKC). In contrast, LTB4 but not cysLT effects are exclusive on syk activation. The aim of this study is to investigate the importance of classical and novel PKCs in LT-mediated phagocytosis and determine the signaling pathways involved. Methods and Results: AMs were pretreated or not with inhibitors of PKC-d (Rottlerin - 6mM), PKC-a (Ro-32-0432-9nM), PI3K (Ly290042-10mM and Wortmannin-10nM), ERK 1/2 (PD98059 - 2mM), cPLA2 (AACOCF3-10 mM), p38MAPK (SB20190-10 mM) and Ca++ (BAPTA/AM-10mM), followed by the addition of LTB4 or LTD4 and challenged with IgG-opsonized red blood cells for 90 min. Our results demonstrate that LTB4- enhanced phagocytosis is dependent on PKC-a, while LTD4 effects are mediated by PKC-d. Interestingly, ERK 1/2 inhibition abolished only LTB4 effects. Furthermore, inhibition of PI3K and Ca++ abolished the effects of both LTs. In addition, we observed that LTB4 induces PKC-a activation before LTD4, while LTD4 activates PKC-d earlier than LTB4. Furthermore, LTB4 activates ERK 1/2 and PI3K in earlier time points than LTD4. Conclusion: We conclude that PKC-a, ERK 1/2, PI3K and Ca++ are essential signaling molecules involved in LTB4-mediated phagocytosis, whereas PKC-d, PI3K and Ca++ stimulate LTD4-enhanced phagocytosis.
