- Abstract: Introduction and objectives: Similarly to apoptotic cells, amastigote of Leishmania amazonenis exposes phosphatidylserine (PS) on their surface (Current Biology 11:1870- 1873,2001). The amount of exposed PS is modulated by the host (J. Immunol. 176:1834- 1839,2006). We quoted this phenomenon as apoptotic mimicry. PS on amastigotes signals for the production of anti-inflammatory molecules, inhibition of NO synthesis, and induction of macropinocytosis. Present objectives are definition of the kinetics of intramacrophagic PS exposure and signaling on the progression of the disease. Methods and Results: In vitro infectivity was assessed by infecting BALB/c and C57Bl/6 peritoneal macrophages, PS exposure by Annexin-V binding, macropynocytosis by lucifer yellow uptake and morphology by confocal microscopy.Now we show that the amount of exposed PS in amastigotes during in vitro infections is the same, regardless of the genetic origin of the host cell. In infected macrophages, the amount of exposed PS in amastigotes increases from 24 to 96 h postinfection, coinciding with the time for fully promastigote-to- amastigote differentiation. Increase in PS exposure correlates with the appearance of large intramacrophagic vacuoles and with a decrease in intracellular proliferation. The signals provided by PS, including lucifer yellow uptake, are down-modulated after parasite internalization and recovered when the intraphagolysosomic amastigotesdisplay maximum PS exposure. Conclusions: PS exposure is a feature of the intracellular promastigote to amastigote differentiation, correlates negatively with intracellular parasite proliferation and our results suggest that it is able to signal from within the phagolysosome.
