- Abstract: Introduction and Objectives: Recently, we showed that nitric oxide (NO) produced during Trypanosoma cruzi infection increases erythrocyte oxidative stress, but, is not necessary for the development of anemia in the acute phase of infection in mice (FEMS Immunol.Med.Microbiol., 41:69-7,2004). Here, we investigated the effects of inhibition of leukotrienes on control of parasite burden and erythrocyte oxidative stressing during T.cruzi infection in C56BL/6 and C57BL/6 iNOS-/- (KO to inducible nitric oxide synthase). Methods and Results: We assessed oxidative stress in erythrocytes from mice infected treated or not with nordihydroguaiaretic acid (NDGA, 50 mg/Kg/mice) by measuring oxyhaemoglobin and haemoglobin derivatives (i.e., methaemoglobin and hemichrome), oxygen uptake and induction time (T ind) following treatment with t-butyl hydroperoxide (t-BHT).We found that the increase of erythrocyte oxidative stress an acute infection of mice with T. cruzi was inhibited by lipoxygenase inhibitor (NDGA) (Induction time:139,7 ± 6,9s vs 171,3 ± 8,3s P< 0,001);(Oxygen uptake: 50,8 ± 2,7 nMOL/mg prot. vs 40,4 ± 3,0 nMOL/mg prot. P < 0,001). Treatment with NDGA significantly increased blood and cardiac parasitism. The heart tissue from treated mice had four-fold more parasites nests than those of non-treated mice on day 12-post-infection (4,1 ± 0,8 vs 1,1 ± 0,15, P < 0.05, Bonferroni multiple comparisons test. The NDGa treatment partially decreasedthe NO production by activated macrophages (45,8 ± 1,5 vs 38,0 ± 1,7, P < 0,001). Conclusion: Taken together, these data suggest that leukotrienes play an essential role in controlling parasite burdens in blood and heart, as well as, in the regulation of NO production from activated macrophages during acute phase of experimental infection with T.cruzi.
