A model of TH1 cell-mediated lung inflammation (2006)

- Abstract: Introduction and Aim: The ovalbumin (OVA) Th2-model of allergic lung eosinophilic inflammation is well established. The aim of the present work was to develop and characterize a polarized Th1-model of lung inflammation. For this, animals were sensitized with OVA plus bacterial lipopolysaccharides (LPS) as adjuvant. LPS is a prototypic TLR-4 ligand and a strong inducer of IL-12 that is essential for Th1 polarization. Methods: BALB/c mice were sensitized with 100 µg of OVA with or without 10 µg LPS, OVA/LPS or OVA group, respectively, by intranasal (i.n.) route for three consecutive days. After sensitization, mice received four OVA i.n. challenges at days 7, 14, 21 and 28, Control group received PBS. One day after the last OVA or PBS challenge. bronchoalveolar lavage (BAL) cell and serum were collected. Results: BAL cells from Control group consisted mainly of macrophages (1.8x105 cells) and lymphocytes (0.2x105 cells). BAL cells from OVA group exhibited a very discrete increase (<105 cells) in the numbers of eosinophils and neutrophils, but not mononuclear cells when compared to Control group. In contrast, BAL cells from OVA/LPS group showed a significant increase in the number of neutrophils (6.5x105), macrophages (7.2x105) and lymphocytes (2.3x105) when compared with Control group. In addition, BAL cells from OVA/LPS group produced high levels of hydrogen peroxide but not nitric oxide. Finally, animals from OVA/LPS group produced high levels of OVA-specificIgG2a, a IFNã- dependent isotype, and higher levels (8-fold) of IgG1 than OVA group. Conclusion: We demonstrate a novel model of Th1-mediated lung inflammation characterized by the influx of mononuclear cells and neutrophils.
