Participação das ß2-integrinas na histoplasmose pulmonar murina (2006)

- Abstract: A histoplasmose é uma doença granulomatosa crônica, cujo agente etiológico é o fungo dimórfico Histoplasma capsulatum. As vias aéreas superiores são rotas para infecção nos mamíferos e a inalação de microconídios e fragmentos de micélio leva a intensa reação inflamatória nos bronquíolos e alvéolos pulmonares. Em pacientes imunocompetentes, na maior parte dos casos, a infecção é limitada ou assintomática. Em indivíduos imunossuprimidos, o fungo pode causar formas pulmonares graves da doença ou se disseminar, atingindo outros órgãos e podendo levar o paciente a óbito. A imunidade celular do hospedeiro determina o grau das manifestações clínicas na histoplasmose, sendo a interação entre células T e macrófagos, fundamental para o controle da infecção e erradicação do H. capsulatum. Dados anteriores da literatura mostram a participação da cadeia 'ß IND. 2' das integrinas, também denominada CD18, em interações entre H. capsulatum e células hospedeiro. No presente trabalho, utilizamos animais geneticamente deficientes para expressão de CD18, para avaliar a função desta molécula na infecção por H. capsulatum. Dessa forma, demonstramos que animais deficientes de CD18 são menos eficientes no desenvolvimento de resposta imune na infecção por H. capsulatum, tendo menor reação de hipersensibilidade do tipo tardio, maior carga fúngica nos pulmões e baço e menor sobrevida em relação a animais selvagens. Nossos resultados demonstram ainda que a deficiência de CD18 resulta emreação inflamatória exacerbada, com recrutamento pulmonar de neutrófilos aumentado em relação a animais selvagens. Através da análise histopatológica de cortes de pulmão, demonstramos que animais deficientes de CD18 apresentam maior comprometimento deste órgão durante a infecção em relação a animais selvagens. Durante a fase tardia da infecção, a produção de mediadores fundamentais na imunidade durante a histoplasmose, como 'LTB IND. 4'. IL-10, TNF-'alfa'., IL-12 e IFN-'gama', está diminuída no pulmão de animais deficientes de 'ß IND. 2'-integrina em relação aos selvagens. Finalmente, determinamos que macrófagos alveolares de animais deficientes de CD18 são menos eficientes para fagocitar leveduras de H. capsulatum. Desta maneira, este trabalho mostra o envolvimento da molécula CD18 na infecção por H. capsulatum, demonstrando sua importante participação durante esta infecção
