Chronic circadian desynchronization decreases the survival of NZB/W(F1) mice (2005)

- Resumo: Introduction: It is generally accepted that organisms derive benefits from the temporal organization of their physiology and behavior. Life expectancy may be reduced for some organisms raised under light-dark (LD) cycles with periods that differ substantially from that of their known circadian rhythm as well as for animals submitted to repeated phase shifts in the LD cycle. Objective: This study assessed the influence of chronic circadian desynchronization on the development of proteinuria and life span in NZB/W (F1) lupus prone mice. Methods: After the weaning, animals were housed in polypropylene cages with food and water freely available and were divided into two groups: experimental and control. The control one maintained on a fixed 12:12h LD cycle, and the experimental was subjected to a 12h phase shift in the LD cycle on a weekly basis. Urinary protein levels were monitored weekly by the use of reagent strips and the duration of life span was recorded for each animal.Results: Chronic reversal of the external LD cycle at weekly intervals resulted in a significant decrease in the survival time in NZB/W(F1) mice, with the median life span being reduced by 21%. Moreover, Student t-test showed significant statistical difference in the interval between the detection of proteinuria and death (p<0,05), and the vatiation coefficient demonstrated greater variability of this interval within the experimental group. Conclusion: Disruptingordinary circadian rhythmicity in animals susceptible to early mortality due to autoimmune disease results in further decrease in longevity.
