- Abstract: Objective: The purpose of this work was to investigate the effect of Aldosterone on Na+- independent mechanism of cellular pH (pHi) regulation in proximal S3 segment. Materials and Results: We investigated the effect of Aldosterone on Na+-independent mechanism of cellular pH (pHi) regulation in proximal S3 segment of rat (Wistar 90g)in vitro, following intacellular acidification by NH4Cl. pHi was mensurad by fluorescence microscopy using the probe BCECF, during superfusion with Na+-free solution. Our results indicated that the mean baseline pHi was 7.08 ± 0.017 (n = 90). In the presence of Na+-free medium, the pH recovery rate following intracellular acidification in control situation was 0.061 ± 0.005 (n = 28) pHi units/min. In the presence of Na+-free medium with Concanamycin (4.6 X 10-8 M; a H+-ATPase specific inhibitor), the pH recovery rate decrease to 0.005 ± 0.007 (n = 24) pHi units/min.In the presence of Na+-free medium with Aldosterone (10-12, 10-10, 10-8 or 10-6 M; 1 h preincubation and superfusion) it increased was 0.16 ± 0.014 (n = 12), 0.13 ± 0.011 (n = 18), 0.13 ± 0.011 (n = 18) or to 0.08 ± 0.030 (n = 21) pH units/min, respectively (p < 0.05). In the presence of Na+-free medium with Concanamycin and plus Aldosterone (10-12 or 10-6 M) it was 0.009 ± 0.008 (20) pHi units/min or 0.008 ± 0.020 (18) pHi units/min, respectively. Conclusion: Our data show that Aldosterone acts on the process of pHi regulation inthe proximal S3 segment with dose-dependent stimulatory effect on the H+-ATPase.
