Gene expression profile of iodide treated rat thyroid folllicular cell lineage PCCl3 (2005)

- Abstract: Objectives: Iodine is one of the main regulators of thyroid functions, which is transported by the NIS protein, localized in the basolateral membrane of thyrocytes. High iodine concentration affect thyroid cell differentiation, decreasing hormone synthesis (Wolff- Chaikoff effect), and inhibit cell proliferation. Although a number of studies have been performed about iodine auto regulation in follicular thyroid cells, the molecular mechanisms of these effects are still not clear. The purpose of present study is to investigate the effect of high iodine treatment on thyroid follicular cells by looking for genes modulated by iodine. Materials and methods: For this analysis we constructed two SAGE libraries from 15ug of total RNA extracted from rat thyroid follicular cell line (PCCl3) cultured in HAMF12 medium with or without 10 - ³ M of iodine for 24 hours. Then, the RNA was reverse transcribed to cDNA and digested with specific enzymes generating 10-14pb tags, which were cloned in pZero vector and transformed in Escherichia coli. Recombinants were sequenced to reach approximately 12,000 tags for each library which were submitted to SAGE® analysis software. Results: The results allowed us to identify 11,897 transcripts, of which at least 60 were up regulated and 40 down regulated in the iodine library. Among them, Smad7, Smurf2, Lgals1, Khdrbs Src, Eif3s3, Ltbp3 and Mapk3 were 8 to 14 times more expressed and NIS , Thyroglobulin, Shc1, Fos1, Adcy6 and Adcy2 were 4 to11 times less expressed. Moreover, 14 genes were expressed only in the iodine library. Conclusion: Differential expression patterns were found for genes already related to and others not yet associated with the iodine auto-regulatory effect. These findings show that iodine excess acts in the regulation of several genes amplifying the molecular spectrum of iodine effect in PCCl3 thyroid follicular cells
