Production and secretion of heat labile toxins by enterotoxigenic Escherichia coli strains isolated from human and porcine hosts (2005)

- Abstract: Enterotoxigenic Escherichia coli (ETEC) represent one of the most important etiological agents of diarrhea in developing countries. Ability to induce disease is characteristically associated with production of colonization factors and at least one of two enterotoxins: the heat-labile toxin (LT) and the heat-stable toxin (ST). Previous studies have indicated that LT produced by ETEC strains isolated from humans (LTh) is distinct of the toxin produced by ETEC isolated from pigs (LTp) with regard to immunological properties, isoeletric point, electrophoretic mobility on SDS-PAGE gel, and amino acid composition. However, the capacity to produce and secrete LTh or LTp by ETEC strains was not previously investigated. In this work we determined the production and secretion of LT in a set of 42 ETEC strains isolated from piglets or humans. The LT concentrations detected in whole cell extracts or in culture supernatants were determined with a sandwich ELISA based on LT-specific antibodies. The LT detected in crude extract of ETEC strains isolated from human hosts ranged from 29.1 to 2,776.6 ng/ml while the toxin secretion ranged from 0 % to 53% of the total toxin produced. On the other hand, the LT levels expressed by ETEC strains isolated from porcine hosts ranged from 9 to 300 ng/ml, while LT secretion varied from 0% to 20% of the toxin produced. These results indicate that the capacity to produce and secrete LT by ETEC strains isolated from human and porcine hosts may varygreatly according to the strain analyzed. Moreover, the present evidences suggest that the natural diversity of wild ETEC to express and secrete LT involves both gene regulation and toxin secretion mechanisms
