Expressão de NAD(P)H-oxidase nas células trofoblásticas (2005)

- Abstract: O Complexo NAD(P)H-oxidase é um doador de elétrons associado a membrana plasmática, enzimas que catalisam a produção de ânions superóxido, espécies reativas de oxigênio (EROS), envolvido no processo oxidativo. Componentes ligados a membrana gp-91-phox, p22-phox citocromo b558 e os componentes citosólicos p47-phox, p67-phox and p40-phox. Recentes trabalhos mostram que células trofoblásticas são capazes de produzir EROS , uma atividade que em humanos pode estar relaciona a problemas gestacionais como a préeclampsia. Entretanto a presença de dos componentes do complexo enzimático foram analisados por imunhistoquímica e Rt-PCR.Métodos: As células trofoblásticas foram obtidas a partir de cone ectoplacentários de embriões camundongo de7,5 dia de gestação. Os cones foram cultivados por 48h, tratados ou não com PMA (100ng/L) de 6-12h e processados para a localização das proteínas e dos transcritos gênicos de gp-91-phox,p47-phox, p40-phox p67- phox todos foram expressos na presença e na ausência de PMA.Resultados: As subunidades gp-91-phox,p47-phox, p40-phox,p67-phox foram foram expressas nas células trofoblásticas na presença ou ausência de PMA. A presença de PMA aumenta a expressão protéica de p47-phox, p67-phox e gp91-phox. As células trofoblásticas expressam as proteínas associadas a atividade enzimática de NAD(P)H-oxidase. Portanto, a expressão destes componentes pode estar sendo modulado por estimulos específicos sugerindo que a NAD(P)H-oxidase pode ter um papeladaptativo na fisiologia da placenta
