Laminin-1 enhances secretion of type I collagen and tenascin in cells derived from human adenoid cystic carcinoma (2005)

- Abstract: Objetivo: Adenoid cystic carcinoma is a frequent malignant salivary gland neoplasm with high level of recurrence and distant metastasis. We have demonstrated that laminin-1 modulates the phenotype of an adenoid cystic carcinoma (CAC2) cell line. We are currently analyzing the role played by laminin-1 enhancing secretory activity of CAC2 cells. Métodos e Resultados: CAC2 cells were grown inside laminin-1, while cells grown within agarose served as controls. Electron microscopy (EM) revealed that CAC2 cells grown within laminin-1 formed pseudocysts filled with secretory-like material. These cells also exhibited prominent and dilated endoplasmic reticulum, and numerous vesicles. Controls cells, grown in agarose, presented spindle-shaped morphology, with Golgi apparatus, myofilaments and scarce basement membrane lamina. These EM findings suggested that laminin-1 induces secretory activity in CAC2 cells. We further investigated this point by light microscopy, immunofluorescence and confocal microscopy. Histochemistry displayed PAS positive material in CAC2 cells treated by laminin. Immunoperoxidase detected type I collagen in these cells. CAC2 cells grown inside agarose (controls) showed no secretory-like material. Confocal microscopy provided additional information on the role played by laminin-1 enhancing secretory activity of CAC2 cells. Whole mount immunofluorescence was carried out in three-dimensional preparations of CAC2 cells grown either within laminin-1(treated) or inside agarose (controls). Optical sections taken 1qm apart showed that laminin-1 enhanced secretion of collagen I and tenascin in CAC2 cells, when compared to controls. Conclusões: Laminin-1 enhances secretion of type I collagen and tenascin in cells derived from human adenoid cystic carcinoma
