Expression of VEGF-B and fgf-b on thyroid hormone-induced myocardial hypertrophy (2005)

- Abstract: Objetivo: The hypertrophy induced by thyroid hormone is well established, however the changes in the cardiac capillaries are still controversial. In our previous studies we found that the hypertrophy induced by thyroid hormone does not change the capillary density. The vascular endothelial growth factor- B (VEGF-B) and the fibroblast growth factor-B (FGF-B) are involved in many physiologic and pathologic angiogenic processes, but few is known about their involvement on hyperthyroidism. Objectives: The aim of this study was to evaluate the VEGF-B and FGF-B expression on myocardium in a model of cardiac hypertrophy induced by thyroxine (T4). Métodos e Resultados: Sprague-Dawley adult rats were treated with 0.100 mg/kg/day of T4, i.p. (T4-100). Body weight and hemodynamic parameters were obtained. After 7 days of treatment, the rats were decapitated; hearts were removed, weighted and processed to analyze the gene expression of VEGF-B and FGF-B by RT-PCR. The increase on myocardial mass (33%) was accompained by an increased VEGF-B expression (4.51 ± 1.02 vs. 0.27 ± 0.17, p< 0.05) and decreased FGF-B expression (0.60 ± 0.37 vs. 1.64 ± 0.40, p<0,05) on the T4-100 group when compared with control group. In the hyperthyroidism there was a balance between the VEGF-B and FGF-B expression, both growth factors frequently involved on angiogenic processes. Perhaps these results might in part explain the absence of alteration on the capillary density in this model of hypertrophy,as have already been observed for us recently
